1. Tube1: 타액 5 ml +1방울의 20% glucose + 20mg of TCP (실험진행)
Tube2: 타액 5 ml +1방울의 20% glucose + 2방울의 toluene (냉장보관)
2. 37℃에서 1시간 30분간 방치
*) 본래 구강세균기질과 타액의 접촉시간이 4시간이상 되는 배양이 필요하나, 실험시간의 제약으로 90분간 배양을 실시
3. 2방울의 toluene을 가하고 4?에서 10분간 방치
4. 8ml의 10% TCA 첨가
5. 300rpm에서 10분간 원심분리
6. 상청액을 희석하여 유리된 P의 양을 정량분석
1) Introduce 1 ml of sample or blank solution into tubes.
2) Add 0.5 ml of 5N sulfuric acid
3) Add 1 ml of acid molybdate reagent
4) Mix thoroughly.
5) Add 0.1 ml of reducing reagent (ANSA solution)
6) Add 2.4ml of DDW
7) Mix well.
8) Read optical density at 660 nm after 10 nm
